Tough choice, but I guess I'd have to go with the big Brothers and Mountain Boys. I jut like boy ponies too much to choose otherwise. Close followups are the So Softs and Twinkle Eyes.

I'm torn between the So Softs, the Rainbow ponies & the original four unicorns. xD
I think the Rainbows would have to be my firm favourites though, there are so many beautiful characters in that set, & I have a soft spot for the pegasai.

Yeah, seriously, I love their unique poses, though I wish the pink one was a pegasus. I also have the full set of 4 ponies in near mint condition (cuddles has perfectly pink hair) so, yeah.

I have to go with the Merry go round ponies. I really love them. I also love the princess brush n grows. this was a hard topic to choose only one with.

I like a lot of individual ponies from a wide variety of styles, and there are certain sets I would like to eventually have all of, but I think my very favorite set is The Perfume Puffs.

The Sweetberry set is second. 
After that I love the original sweetheart sisters, the princesses, and sea ponies.
