Good question!

My main, longstanding grail was obtained thanks to hathorcat, who traded me the most beautiful FF Minty (the pony I originally wanted back in 1984 - she was impossible to find, so I'd settled for Blue Belle, and thus began my collecting ).

My secondary one was Night Glider, and I got her from Emberfly.

Now I'm on to my tertiary grails, which are (in no particular order):

Hip Holly
Winter Minty
Candy Cane
Dream Drifter (on her way from Jinxxy! )
Flim & Flam blindbags (hurry up, next wave of blindbags!!!)

But I'm happily obtaining other ponies of all sorts, including some that would probably have been grails had I known about them before getting my hooves on them.

My grail was Macau Jenny, but I managed to find her. There are some ponies that I still want, but not as badly as I wanted Jenny!

Just recently I've obtained the closest thing I've had to a grail since I've started collecting, G4 Minty Type A! 

The next hard to find pony I'd really love to have is Sweetie Blue. I think she's going to be much harder to get though

Mmm, non-Nirvana is Red Roses for sure.  I have a Mimic, though.

Nirvana?  German Starshine for sure.  She's in Surprise pose, which is my absolute favorite, and I need to increase my Surprise pose army!  I'm most sad that pose never came in TE OR a perfume puff.  ;.;

I don't have a grail, but I'd love to get my hands on some G1 Twinkle-Eyed (MOC) and the Merry Go Round (MIB) ponies.

no Grail at the moment,  but would love to get my hands on some G2's but not willing to go past 30 dllrs for them and they keep going up and up , 80 the last time i was watching them on ebay , the boat of the G2 is another item i would like to get someday....

No pony grails for me right now - just accessories. And right now it would be the little minty green ribbon barrette which came with the Great Hair ponies.
