Well, with the power of the Internet, most ponies are attainable. 

For secondhand market, I would say most of the mail order babies.
For Nirvana, I would say a complete toyline of G1 Takara products.

Oooh, that's a good one, Lovesbabysquirmy. I'd love a Takara pony, but there's only been, what, 3 sighted? And I don't wanna pay $1000 for a pony, LOL!

I'd also love (but will never get) some unreleased pony prototypes, like the set of "Molly's friends" who came with pony friends!

Well there's holy grails that cost more than my both of my paychecks in a month, but they're not really my most wanted. I'd say a lot of G3's that I like but not because of rarity but more because of my money constraints and shipping costs. For dolls though, I honestly feel as though I'll NEVER get Cedar Wood.

Mimic, Munchie, Baby Princess Sparkle ( this one maybe one day but not anytime soon) , Blueberry Baskets...any of the harder to find uk variants ( big brothers especially), the harder to find so soft ponies with full clean soft fuzz (Twilight and the pretzel girl in particular) xD

OH and all the TAF babies. SO much want, but not getting xD

For me, it's finding the last few ponies on my list at decent prices. For some reason, people like to jack up the price of individual ponies from sets so that the one pony itself costs almost the same amount as the set it came from, or even higher *cough* CMC & Friends *cough*

TRU , Silver Spoon, Twist-a-loo, Star Dreams, and Costco's Minty are some of the ones I am in need of.

I'd be careful before deciding that you'll "never get" ponies that you really like.  Back when I first started collecting years and years ago, I was convinced that I'd never own the likes of Mimic or SS Crumpet or Rapunzel, but slowly, even those HTF/highly sought after ponies that I'd imagined being impossible to find have made it into my collection.  If they really are that special to you, keep looking and they'll show up at the right time and right price eventually

For what it's worth, the ponies that I'm actively seeking right now that I think I'll have the hardest time finding include several G1 mail orders (babies, Mommy and Baby, Scribbles and Dabble...) and G3s Licensing Show Pinkie Pie and both Breast Cancer Awareness Ponies.  They're on my watch list, but I'm thinking I'll be holding out for the long haul on them.

Spain 4-Speed and Salty. They're just a bit too expensive for me, as much as I love my BB ponies. Who knows, maybe in the future I'll be in a position to get one, but it seems unlikely.
