I was examining a book I found thrifting the other day, kind of perplexed with it- I had never heard of it before finding it, and could find no documentation of it online. It's a Play-a-Sound book called "Hop, Skip, & Sing!" with Twinkle Twirl on the cover. Her symbol isn't visible, and neither are some other ponies', so I was going off of color scheme, hoof heart, and eye detail to make sure my identifications were correct. Then, Twinkle Twirl's eyes stood out to me...
Okay, so every G3 has two highlights in her eye: one large white one, and a smaller colored one. The smaller one generally comes in three shapes: a circle, heart, or five-point star. Twinkle Twirl does not have any of those three shapes; in fact, she has two small highlights! How many other ponies break the norm?
I know of three or five, depending on whether you count G3.5.
Applejack has an apple. I used to think it was a heart until I got mine- I noticed while inspecting her "smudged" eye!
Twinkle Twirl has a white four-point star with a small reddish-pink circle next to it. As far as I know, she's got the only "double" highlight.
Waterfire has a flame. Hers is my favorite!
Scootaloo had a heart in G3, but in G3.5, it's a butterfly.
Cheerilee had a flower replace her star in the transition to G3.5.
In addition to these, each pre-Newborn So Soft (Rose Blossom and friends) has a more detailed highlight which is a mini version of her symbol.
Are there any others? There are a lot of ponies I don't have, and this detail tends to be quite small in photos.

Most of my G3 are in storage, and the ones that live down here with me fit into your categories of circle, star or heart :)

Checked my G3s and didn't see any others than those you mentioned! I checked my babies, too.

Waterfire's little "I'm gonna watch it all burn" flame is the most amazingly hilarious thing to me. She's a wild child, that one.

I looked for ponies with extra features or design elements, and the only other one I found was the never-officially-released Ribbons & Hearts. Hers is shaped like a ribbon, and is quite detailed too!
On an unrelated note, did you know that they redesigned Twinkle Hope? She had a costume made for her like the ones for The World's Biggest Tea Party. She apparently was at the Give Kids the World Village until as late as 2017! They replaced her with a G4 version without eye/mouth animation and solid hair. She looks less like she could eat souls, though, so that's a plus!
