No, the cards are their own separate thing. They come (in NA) as packs of six with 5 regular cards and one special / shiny card. The figures are going to be released around the time that Animal Crossing Amiibo Festival is. If I remember correctly, two are coming with the game (Isabelle and another I can't remember), three are being sold as a 3 pack, and then there are a couple (Tom Nook + again memory is failing here) that are being sold as single figures. The cards work the same in function as the Amiibo figures do, they're just cards instead. Hope that helps.

I don't think it really matters if an amiibo card has been used or not, or at least I have yet to hear anyone say "my friend tried to use my ____ card, and the game wouldn't let her". Almost certain that the cards are reusable.
Plus I think if you trade cards with someone and they already used the card, you can get furniture out of it and/or see how that person designed that character's house (I think you have to save/update the card in the game? Something like that).
I remember reading something about it on reddit - I'll try to find it.
